Cellular proliferation and structural cromosomal aberrations in cells submitted to the action of Brazilian dolomita (Gran-White) in vitro

The Brazilian dolomite (Gran White) is a mineral alimentary supplement used as adjuvant in the prevention and treatment of the osteoporosis, because it increases the bone mass. This investigation was accomplished with the purpose of analyzing Brazilian dolomite action in cells obtained by conventional culture of lymphocytes with relationship to the occurrence of chromosomal aberrations, micronuclei and to the mitotic index. Study of case-control was made in composed sample by 2 elders, 2 younger seniors and 2 students in the 20-30 year-old group. The study in vitro was accomplished with application of the substance in 3 different concentrations (1,0mg, 3,0mg and 5,0mg), besides the historical, negative and positive controls (without substance increment, with Vitamine C and with ciclofosfomide, respectively). In each treatment 50 cells were analyzed in common optical microscope. The results showed action of dolomite in the increase of the mitotic index on the experiments, in concentrations corresponding to the administration in vivo. However, the spontaneous rate of micronuclei and the chromosomal changes had significant increase in the different doses of the substance in seniors, and a smaller proportion in the youngest. The obtained results show the positive effect of Gran White in the rate of cellular reproduction, agreeing with clinical descriptions of increase of the bone mass. However, it could not be attributed to the substance a protecting effect against clastogenic agents.Downloads
