Abstract

Mesenchymal stem cells show excellent characteristics for use in cell therapy. However, the extraction, maintenance andexpansion protocols of these cells need to be standardized. The aim of this study was to assess the influence of the pre-cultureperiod (PCP), trypsinization time (TT) and addition of conditioned medium (CM) on the yield of a primary culture of mesenchymalstem cells extracted from the bone marrow of mice. Our results show that the groups of cells cultivated for a PCP of 72 hours hadbetter cell yield, compared to a PCP of 24 hours. The TT showed no significant difference between 5 and 10 minutes; however,cell viability remained above 90% for both times assessed. The conditioned medium added to the primary culture seems to haveno significant influence on cell yield. We conclude that the use of a PCP of 72 hours significantly increases the number of cellsobtained in a primary bone marrow culture.
