Evaluation of the micronuclei from inflammatory cells into sporotrichosis and chromoblastomycosis patients

Abstract

Subcutaneous mycoses as the sporotrichosis (ST) and chromoblastomycosis (CM), possess great incidence in Latin America. Their clinical signs happen initially with the formation of nodular lesions in ST and verrucoid or warty lesions in the CM, both developing for granulomatous microabscesses merged with pseudoepitheliomatous hyperplasia, that consist of neutrophils, macrophage and Langhans’giants cells. As consequence of that immune response, the inflammatory cells intensify theirmetabolism and increase the protein synthesis, what leads the micronucleus formation. Micronuclei are chromatin fragments proceeding aberrant mitoses. The aim of this study was to assess the frequency of micronuclei of inflammatory cell in the cutaneous lesions of ST and CM. Biopsies of epidermal tissue of patient carriers of ST (n=10) or CM (n=10) were selected. Cytochemistry was carried out by staining with Feulgen reaction and the counter-stained with Fast Green. The obtained results indicate that micronuclei frequency is significantly higher in the characteristic lesions of ST, when compared to the cases of CM (p < 0.05). The preliminary results suggest different cytogenetic mechanisms of response of the inflammatory cells, although the histopatologic profiles of the lesions are similar.
